BACKGROUND

FIGURE 5: NONSPATIAL WATERMAZE A rat is placed in the pool from a constant start position. The location of the submerged platform varied but was always associated with an intra-maze cue (vertical lines). The rat was not reliant on extra-maze cues to locate the platforms. Each rat received 4 test trials a day and was tested for 4 days.

• Hypothermia has not been examined in preterm infants due to concern for co-morbid health issues. Caffeine is routinely used in preterm/ELBW and VLBW infants and been shown to effective in decreasing apnea, cumulative amount of hypoxia and secondary outcomes including cognitive outcomes.

FIGURE 3: ROTAROD This paradigm measures how long a subject can stay on a rotating rod, with maximum acceleration after 5 min. The time each rat could stay on the gradually accelerating rotating rod was recorded in seconds.

• However, hypothermia has not been trialed in preterm infants, due to concerns regarding co-morbid health issues. In preterm/VLBW infants, caffeine therapy has been effective for treating apnea of prematurity, and has secondarily been found to improve rate of survival.

AIM OF THE STUDY

• The current study sought to investigate the potential therapeutic effects of caffeine citrate and hypothermia using a preterm (P6 HI) rodent HI model.

METHODS Subjects : Male Wistar rats (n=64) born to time-mated dams (Charles River) at the University of Connecticut were randomly assigned to one of two groups (sham or HI) within litter. These sub-groups were further assigned to normothermic or hypothermic conditions during/after surgery, and to saline or caffeine treatment. Subjects: Data is reported here is for experimental groups Shams, HI-saline-normothermia, HI-caffeine- normothermia, HI-saline-hypothermia.
