Tips and hints

1. *Blanking Solution ST 1600/2000 is used as on-line cleaner to rinse the WBC counting chambers, RBC Plt counting chambers and both metering tubes. 2. ProClean CD is not advised for use on CD 4000. This analyser should use Hypochlorite solution. 3. CyMet 1600/2000 is not suitable for use on CD 1200, using it will result in incorrect WBC Diff results. CD 1200 are no longer in production. 4. For use on the CD 3200 Leucolyse, nr. 3827.5000PC, is needed to count WBC’s and CyMet 3200, product nr. 3823, to measure the Hemoglobin. Leucolyse, nr 3827.5000PC, has the same application as the Abbott product CD3200 WBC Lyse. CyMet 3200, nr 3823, is equivalent to Abbott product CD3200 HGB/ NOC Lyse.

5. J.T.Baker’s calibrator CalSet can be used on CD 1600/1700 to calibrate the standard non diff parameters (WBC, RBC, HgB, MCV, Plt, MPV). It cannot be used to calibrate diff values. Differentials cannot be calibrated on CD 1600/1700. CalSet cannot be used on CD 3200/3500/3700/4000. These instruments use a different measuring principle. 6. 8 Parameter Control available in 2.5 ml, 4.5 ml pierceable and 8 ml vials in three levels. 8 Parameter Control cannot be used on 5 part diff analyzers. It does not work as non-diff control on these analyzers. It is recommended to participate in J.T.Baker’s free external/internal quality assessment program Rapid Stat. It is always recommended, in case of changing reagent type or reagent origin, to check whether a re-calibration is required.

Cleaning of Abbott’s automated Cell-Dyn analyzers

As most end-users know, a regular detergent is used on the automated Cell-Dyn analyzers. This detergent is colorless and is meant to clean the hemoglobin flow cell every sample. The CD3000 and CD3200 are exceptions, since these ones don’t require a detergent on-line. The Mallinckrodt Baker on-line detergent is the Blanking Solution ST1600/2000 (3947). What a lot of end-users don’t know is the use of an additional cleaning on the Cell-Dyn analyzers. This cleaning step is a necessity to clean the whole analyzer, preferably once a week. Selecting from the

MAIN MENU should activate this cleaning step by proceeding next steps: 1. SPECIAL PROTOCOL 2. MORE 3. AUTO-CLEAN or EXTEND AUTO-CLEAN Follow the steps as displayed on the computer screen. Use for this special cleaning ProClean CD (3902) to fulfill a sufficient cleaning of the analyzer. With this cleaning procedure the whole analyzer is thoroughly cleaned.

Priming and Calibration of Abbott CD Analyzers Abbott CD1300 - CD2000 This priming protocol can be used for several different Celldyns: CD1300, CD1400, CD1600, CD1700 and the CD2000. Priming of Baker reagents as described in the protocol is necessary because these Celldyn instruments have an internal reagent reservoir for the diluent and several syringes filled with Abbott reagents. Before switching from original reagents to Mallinckrodt Baker reagents it is important to know that the instrument is running without any problems and that the values measured for the blood controls (all three levels) are within the specifications of the assay value sheet. Therefore it is necessary to run blood controls or some fresh reference patient samples before you switch to our reagents. These values are necessary for the calibration afterwards. Priming of Cell-Dyn 1300/1400/1600/1700/2000 1. Take the aspirating tubes from the Abbott reagent cubitainers. Don’t lay the tubes on tissues or dusty surfaces. Place them in clean beakers.

2. It is important to rinse the outside surface of the aspirating tubes with distilled or de-ionised water to be aware of any kind of reagent contamination. 3. Press key. 4. Then press key. Press two times . 5. Press to prime isotonic diluent and detergent/blanking solution. 6. Then press . 7.

Put the aspiration tubes into the cubitainers of Blanking Solution ST1600/2000, Diluid ST1600/2000 and CyMet ST1600/2000.

8. Repeat step 5 at least 4 times and step 6 at least 6 times. 9. Press , to shut down the instrument.

12. When auto-startup prime cycle and background check are complete, review displayed data. Acceptable values are as follows: Background Count Time: WBC < 0.5 K/µl 4.50 + 0,20 s RBC < 0.05 M/µl 7.00 + 0,20 s HGB < 0.2 g/dL PLT < 10 K/µl 13. When acceptable background values are displayed the priming is successful. If not, repeat point 5 and 6 again. Calibration of Cell-Dyn 1300/1400/1600/1700/2000 FOR CALIBRATION INSTRUCTIONS AND CALCULATING NEW CALIBRATION FACTORS REFER TO THE PARAGRAPH GENERAL INFORMATION.

Choose the option from the MAIN menu. The calibration menu displays the calibration factors for the open mode. Enter new calibration factors by pressing . Enter the new factors by using the numeric keys. Go on to next factor using key . Note: There are CellDyns in the market what have a closed mode system. To change the calibration factors for this closed mode you have to enter a hidden screen. This hidden screen contains so called dilution factors for the open and closed mode. These dilution factors have the same function as calibration factors and are calculated the same way as calibration factors. To enter this hidden screen enter 94043 from the calibration menu. The hidden screen will be accessed and the factors are shown. These dilution factors for the open mode are also very useful when you have calculated a calibration factor that is outside the acceptable range. It is then possible to change the dilution factor instead of the calibration factor. Leave the hidden screen by pressing the key.

10. Press on and then on .

2. Leave the calibration menu by pressing the key. Go to the main menu with the key.

11. The system will be primed and background counting is performed.

3. It’s also possible to run the calibration from the AUTO-CALIBRATE option.

4. The internal reagent reservoirs on the left side of the instrument will be emptied. Be sure that no reagent is left in these reservoirs. 5. DO NOT place the aspiration tubes in the cubitainers with J.T.Baker’s reagents yet. Press on the keyboard . The reagent left in the aspiration tube is aspirated into the reagent reservoirs. 6. The instrument will give a FAULT alarm. Place the aspiration tubes into the cubitainers with J.T.Baker’s reagents. Press . All J.T.Baker’s reagents will be aspirated into the reagent reservoirs. Note that there is some mixing up of Abbott and J.T.Baker reagents in the HGB and SHEATH reservoir. 7. To prevent any problems of contamination in the instrument, empty the internal reagent reservoirs again by pressing . Then fill the reagent reservoirs by pressing . Repeat this step at least 3 times 8. Return to the menu and choose . Perform at least 3 blank counts and check the background values. Acceptable values are:

1. Choose from the MAIN menu. The calibration menu displays the calibration factors for the mode indicated. With the functional key you can switch between the open and closed mode calibration factors. Printout the factors for both modes by pressing the PRINT SCREEN key on the KEYBOARD.

Note: The CD3000/3500/3700 use calibration factors and dilution factors. These dilution factors have the same functions as the calibration factors and are calculated the same way. It is possible that the new calculated factor is falling outside the range of the calibration factor. In this case you have to change the dilution factor. If your new calculated calibration factor is outside the range go to step 2 otherwise go to step 3. 2. Press F12 and then F1 on the keyboard. A screen with DILUTION FACTORS for the open and closed mode appears. Enter the new factor for the parameter that has to be calibrated by using the numeric keys. Go to the next factor you want to change by using the arrow keys. When you change the open mode dilution factor it is wise to directly change the closed mode dilution factor again for the parameter you changed. A change in reagents will give the same difference in open and closed mode. Leave the dilution factor screen by pressing the functional key . Go to step 4.

3. Choose from the main menu of the software program by using the functional keys. Then choose to empty all reagent reservoirs on the CD3000 and press for each separate reagent reservoir on the CD3500 and CD3700.

Priming of Cell-Dyn 3200 1. Remove the aspirating tubes from the Abbott reagent cubitainers. Don’t lay the tubes on tissues or dusty surfaces. Place them in clean beakers.

2. Rinse the outside surface of the aspirating tubes with distilled or de-ionised water to prevent any kind of reagent contamination. 3. Choose the option from the main menu of the software program by using the functional keys. Then choose . Choose the functional key from the reagent you want to replace: Abbott Diluent/Sheath HGB/NOC lyse WBC lyse

Mallinckrodt Baker Diluent/Sheath 3200/4000 Cymet 3200 Sheath Fluid 3000/3500

For example press to replace the HGB/NOC Lyse for Cymet 3200. 4. The Special Protocol function will be activated and it will take some time to empty the internal reagent reservoirs or the reagent lines from the reagent selected. Wait until the special protocol has finished and the function key toggles to . 5. Place the aspiration tube in the cubitainer with J.T.Baker reagent. Press on the keyboard . The selected reagent will be aspirated into the reagent reservoir or into the reagent lines. REPEAT STEPS 3 TO 5 FOR ALL THE THREE REAGENTS CONNECTED!!

6. The reagent reservoirs and lines are now filled with J.T.Baker reagents. But there are still some Abbott reagents left in the flow panel tubings and the syringes! To remove this we choose the option from the main menu. Then choose and . A shutdown cycle will be initiated. When the shutdown cycle is finished the message STANDBY will be displayed in the Status Box. 7. When the instrument is in Standby press the functional key . The instrument will perform a Prime cycle and will come into the READY state. 8. Perform at least 3 blank counts and check the background values. Acceptable values for the Background are:

Note: When changing from Abbott reagents to J.T.Baker reagents a recalibration for the hgb is required. Calibration of Cell-Dyn 3200 Analyzers FOR CALIBRATION INSTRUCTIONS AND CALCULATING NEW CALIBRATION FACTORS REFER TO THE PARAGRAPH GENERAL INFORMATION.

1. Choose from the MAIN menu. The calibration menu displays the calibration factors for the mode indicated. With the functional key you can switch between the open and closed mode calibration factors. Printout the factors for both modes by pressing the PRINT SCREEN key on the Keyboard. Note: For the CD3200 it is wise not to change the calibration factor for the HGB but the dilution factor. It is possible that the HGB shows a difference from more than 1.5 mmol/L or 2.4 g/dL. The dilution factors have the same functions as the calibration factors and are calculated in the same way. It is possible that the new calculated factor is outside the range of the calibration factor. In this case you have to change the dilution factor. If your new calculated calibration factor is outside the range go to step 2 otherwise go to step 3. 2. Press F12 and then F1 on the keyboard. A screen with DILUTION FACTORS for the open and closed mode appears. Enter the new calculated factor for the parameter that has to be calibrated by using the numeric keys. Go to the next factor you want to change by using the arrow keys. When you change the open mode dilution factor it is wise to directly change the closed mode dilution factor again for the parameter you changed. A change in reagents will give the same difference in open and closed mode. Leave the dilution factor screen by pressing the functional key . Go to step 4. 3. Choose . You will now see a screen with both the calibration factors for open and closed sampler. Enter the new factor for the parameter that has to be calibrated by using the numeric keys. Go to the next calibration factor you want to change by using the arrow keys. When you change the open mode calibration factor it is wise to directly change the closed mode factor again for the parameter you changed. A change in reagents will give the same difference in open and closed mode. 4. Leave the calibration menu by pressing key . Go to the main menu by pressing the functional key . 5. It’s also possible to run the calibration from the AUTOCALIBRATE option. Refer to the operator manual for this procedure.
