Automating HPLC without Complications

Tasks Requiring the Operator’s Attention

Industry-Leading Column Chemistries

To meet your LC objectives, Intelligent LC delivers “ease of use” by providing specific automation advantages. The UltiMate 3000 is a selfsufficient HPLC—a system that performs with less operator effort. Prepare the system—fill solvent reservoirs, load sample trays—and the UltiMate 3000 runs on its own. System startup and equilibration are equally simple. On-line degassing and purging can be executed automatically through a split-loop autosampler. A single software command primes the autosampler syringe to ready the system for operation. Most importantly, this level of automation does not require much of your time. Running a system by doing almost nothing—that is “ease of use.” This level of automation frees you to focus on what really matters: results.

As far as maintenance, tasks requiring user input are reduced to a minimum, and the remaining procedures are simple enough to be completed within minutes. Maintenance access through the front door of each module is unrestricted. Degasser assemblies, outlet frit, pump heads, injection valves, switching valve, columns, flow cells and lamps are accessible with minimal effort. Special tools and special procedures have been eliminated to ensure convenience and simplicity.

Take advantage of Dionex’s expertise in column chemistries to simplify analysis and address all of your separation problems. The UltiMate 3000 series is configurable for your application using the Dionex Acclaim®, ProSwift™, PepSwift™, or ProPac® column.

Acclaim Silica Reversed-Phase Columns Ultrapure silica, robust bonding chemistries and exhaustive end-capping has created columns with exceptional stability, reproducibility, efficiency and peak symmetry for acidic, neutral, and basic analytes. Acclaim reversedphase columns deliver superior separations for a wide range of analytes, including small molecules, proteins and peptides, organic acids, surfactants, and explosive compounds. Monolithic and Polymeric Columns Dionex’s ProSwift and PepSwift monolithic supports and ProPac polymeric particles offer valuable alternatives to the classical microparticulate sorbents. The monolithic medium is a continuous, rigid, polymeric rod with a porous structure that delivers fast, high-resolution separations. For polymeric phases like ProPac IMAC, the high surface area and inertness of micro-pellicular beads delivers analytical efficiency far superior to traditional capture-release affinity phases.

Intelligent LC solutions, such as automated application switching, provide extensive automation to laboratories. Even when two significantly different methods are required, UltiMate 3000 can provide outstanding results for both separation applications. One instrumental set-up alternates between the methods, unattended, greatly increasing productivity and convenience, and maintaining regulatory compliance. That’s Dionex x2 LC capability.

Compare plates/meter. Check peak symmetry and column stability. These columns have earned the name “Acclaim.” Dionex also offers semipreparative columns with consistent performance that simplify scaling up to semipreparative separations.

Enhanced Reliability Dionex Ensures the Utmost Confidence in Instrument Operation

In designing the UltiMate 3000 series’ high-precision instrumentation, Dionex carefully tested prototypes and evaluated data on component mean-time-beforefailure. We then refined our designs and updated assembly techniques, culminating in extensive testing at the component and system level. To ensure reliable flow rates, our manufacturing and design implementation was evaluated down to the microscopic level. As a result, the surfaces of the new sapphire pistons, piston seals, check valves, and flow cell windows ensure flawless operation at higher system pressures.

Reliability in the Design Reliability was the primary goal for numerous developments in the UltiMate 3000 series. Advances are built in to the design of every module. • • • •

New hydrophilic frits in the solvent organizer reduce the formation of bubbles. A new on-line vacuum degasser enhances the reliability of proportioning, pumping, and detection. Refined flow paths and machining techniques for the pump heads ensure rapid clearance of bubbles that are accidentally drawn into the pump. A new floating piston (see illustration) corrects for imperceptibly small alignment issues that shorten seal life.

• •

A second fan in the variable wavelength detector optimizes the lamp’s thermal environment for life times beyond 2000 hours. Tested flow cells guarantee leak-tight operation up to 100 bar.

The goal of superior reproducibility is achieved with the UltiMate 3000. By ensuring quality parts and performance, Dionex sets a new standard for HPLC reliability, delivering the utmost confidence in instrument operation, analysis, and results.

Seal life is extended by a new floating piston design. The piston’s ability to float and self-adjust corrects for minute alignment imprecisions.

An active seal wash system maximizes seal life for high salt mobile phases. Active monitoring of seal tightness detects leak rates as low as 30 µL/h.

page 6

Confidence in Self-Tests, System Backup

Easy Maintenance with Intelligent LC

Even with the best system components and design, LC reliability greatly benefits from backup mechanisms. UltiMate 3000 pumps eliminate abrasive wear from salt crystals using the industry’s most efficient seal wash system—a standard feature on every pump. Ensuring even greater confidence, automatic monitoring of piston seal tightness alerts the operator to minute leaks before pump accuracy and precision are compromised. Advanced leak detectors deliver reliability in all modules by protecting against solvent exposure.

Since the best maintenance is preventive maintenance, UltiMate 3000 modules and Chromeleon instrument control work in concert to support a timely, cost effective maintenance plan. Autosamplers track injections for the syringes and valve seals. Detector lamps include ID chips that monitor aging. Each of these self-diagnostic features is backed by dozens of

other diagnostic tools. Chromeleon consolidates this information, tracks status, and alerts the operator to take corrective actions. Unexpected failures and repeat assays are avoided, ensuring maximum uptime and reliable operation.

The lamp compartment is thermally isolated from the optical bench of the VWD-3000 detector. The temperature of the compartment is separately regulated by a second fan. By this careful design and continual self-checking, the detector optimizes operating temperature, ensuring a stable baseline for accurate integration of even small peaks.

A qualification status monitor tracks system status and alerts the user of pending requalification.

Increase Throughput The UltiMate 3000 Improves Speed and Productivity

A common goal in chromatography is to separate all peaks in as short a time as possible. One approach is to use columns with smaller particles (3 µm or less). However, when working with particles below 2 µm, the back pressures involved increase significantly. This pressure increases the stress on wear parts, creating challenges for operation—leakage and unwanted extra column volumes, increased instrument down time and reduced chromatographic performance. The UltiMate 3000 Rapid Separation LC (RSLC) delivers a well balanced solution to meet today’s LC performance requirements, provide compatibility with conventional and ultrahigh performance LC columns, and assist users to optimize existing methods to improve analysis speed and peak resolution. Automated Method Scouting is another tool to increase the efficiency of time-consuming method development tasks. It screens up to six columns under different elution conditions and presents the best methods for your application instantaneously.
